New technology offers fighting chance against grapevine killer — ScienceDaily

When the half-inch-long flying insect feeds on grapevines, it transmits bacteria that causes Pierce’s Disease. Once infected, a vine is likely to die within three years — a growing problem for California’s $58 billion wine industry. Currently, it can only be controlled with quarantines and increasingly less effective chemical sprays.

New gene-editing technology represents hope for controlling the sharpshooter. Scientists at UC Riverside demonstrated that this technology can make permanent physical changes in the insect. They also showed these changes were passed down to three or more generations of insects.

A paper describing the team’s work has been published in the journal Scientific Reports.

“Our team established, for the first time, genetic approaches to controlling glassy-winged sharpshooters,” said Peter Atkinson, entomologist and paper co-author.

For this project, the researchers used CRISPR technology to knock out genes controlling the sharpshooters’ eye color. In one experiment, they turned the insects’ eyes white. In another, the eyes turned cinnabar, a blood-red color. Then, the team demonstrated these eye color changes were permanent, passed along to the offspring of the modified parents.

CRISPR is based on the immune systems of bacteria. During attacks by viruses, bacteria save pieces of DNA from their invaders. When the viruses return, the bacteria recognize, cut, and destroy the viral DNA.

Scientists use CRISPR like “molecular scissors” to target specific DNA sequences.

“This is a great technology because it can be so specific to one insect, and not cause off-target effects on other insects, animals or humans,” said Inaiara de Souza Pacheco, UCR entomologist and lead study author. “It’s a much more environmentally friendly strategy for insect control than using chemicals.”

One of the interesting discoveries the team made is that sharpshooter eye color genes are located on non-sexual chromosomes. All animals have two types of chromosomes: sex and autosomal, or non-sexual.

“Knowing that white and cinnabar genes are on autosomal chromosomes demonstrates that the inheritance of these genes is not related to the gender of the insect,” Pacheco said. “This is important for developing control strategies.”

For example, in mosquitoes, it is exclusively the females that transmit viruses to humans. Identifying genes on sex chromosomes that favor female mosquitoes is important for mosquito-control strategies. Conversely, it’s important to know when key genes are not on sex chromosomes.

To demonstrate that CRISPR-made mutations pass through to subsequent generations, the team also had to establish how to get the sharpshooters to mate in pairs. “That’s not always straightforward in entomology, because insects sometimes need more than one other insect to get stimulated for mating,” Atkinson explained.

Now that the team has demonstrated that CRISPR can work in these insects, they have a new goal.

“We’re using CRISPR to try and modify the mouth parts of the sharpshooter so they can’t pick up the bacteria that causes Pierce’s Disease,” said Rick Redak, UCR entomologist and paper co-author.

There is high likelihood the team will succeed in modifying the mouths, given the efficiency with which they were able to change the genes for the sharpshooters’ eye color. The team injected the CRISPR molecules into recently laid eggs, and in some experiments as many of 100% of the eggs became nymphs with altered eye color.

“It’s absolutely amazing because the success rate in other organisms is often 30% or lower,” said Linda Walling plant biologist and paper co-author. “The high rate of gene editing success in glassy-winged sharpshooters bodes well for our ability to develop new methods of insect control, as well as understanding the basic biology of this devastating pest.”

Atkinson also marvels how close the team is to its goal of creating insects that aren’t infectious. “Before CRISPR, generating specific mutations with such ease at such high frequencies was virtually impossible,” Atkinson said. “Now we are confident we can come up with ways to create insects unable to transmit this disease.”

“The outcome of this research is an example of the strength that the agriculture departments in UCR’s College of Natural and Agricultural Sciences bring to developing innovative pest control strategies,” he said.

In addition to scientists from UCR’s Department of Entomology, the research team included Walling from the Department of Botany & Plant Sciences and mycologist Jason Stajich from the Department of Microbiology & Plant Pathology.

Their work was funded by the California Department of Food and Agriculture, as well as the U.S. Department of Agriculture’s Animal and Plant Health Inspection Service.

The team is particularly encouraged by the results of their CRISPR experiments on sharpshooters, part of a class of insects for which other molecular control strategies have not previously been effective.

“It’s looking like sharpshooters will become a model organism for the Hemiptera, this big category of piercing, sucking insects,” said Redak. “Our model of using CRISPR for them could blow open our ability to control diseases they transmit to plants and possibly, to humans as well.”

🏀WBB: Chadron State’s upset bid of ThunderWolves falls short in OT

Zach Carattini, CSC Sports Information

The Chadron State College women’s basketball team (7-13, 6-9 RMAC) lost 77-74 Saturday night in a close overtime clash with Rocky Mountain Athletic Conference rival CSU-Pueblo (18-5, 14-3 RMAC).

Against one of the strongest defenses in the conference, the Eagles kept their composure and played hard for five periods. It was missed free throws and a critical steal by the Thunderwolves late in overtime that sealed the loss for Chadron State in the end.

The Thunderwolves scored the first points of the game off a three-pointer by Ro Dominguez. CSC would tie it up with a free throw by Olivia Waufle and a layup by Samiyah Worrell. Immediately answering back was Pueblo’s Morgan Dial who sank a three-pointer of her own. From there the Thunderwolves would pull away, finishing the quarter with a 19-11 lead over the Eagles.

After allowing a layup and a three-pointer to start the second quarter, the Eagles began to fight back as Jori Peters and Olyvia Pacheco each swished a three-pointer while Bailey Brooks landed two jumpshots and a layup to help make it 35-32 Thunderwolves at halftime.

It was a back-and-forth fight in the third quarter until Peters stole the ball from Pueblo’s Jazzy Hughes and swished a fastbreak three-pointer at the top of the key to make the score 49-47 Thunderwolves. After a missed layup by Alisa Davis, Ashayla Powers tied up the game off a layup assisted by Peters.

Giving the Eagles their first lead of the day was Powers, who put up another layup in the paint to make it 51-49. The Thunderwolves quickly put up two more field goals to gain back the lead right before the fourth quarter. The score was 53-51.

CSC swiftly flew to a five-point lead in the fourth quarter after Peters shot a three-pointer and a free throw followed by a three by Pacheco to make it 58-53. Then Pueblo around and gained their own five-point lead at 2:56 due to four turnovers and three fouls by the Eagles.

With the scoreboard reading 66-61 Thunderwolves, Chadron State scored five unanswered points all from free throws by Pacheco, Powers, and Brooks. At 0:27 on the clock, Pueblo would miss a jump shot and a last-second three-pointer to send the game into overtime with the scores tied at 66. T

The Eagles made only nine of their 16 free throws in the fourth quarter.

Chadron State initially took a 71-68 lead to start overtime. After both teams went back and forth for a few minutes, the Thunderwolves’ Alisha Davis tied the score at 74-74 off a layup with 14 seconds left.

CSC quickly called a timeout with 11 seconds on the clock. Powers passed the ball inbounds but was intercepted by Pueblo’s Amaya Lewis, who sprinted down the court to land the go-ahead layup with seven seconds remaining.

After another timeout, the Eagles turned the ball over again when Worrell tried to pass the ball inbounds but it flew over the reach of her teammates’ hands and went out of bounds.

Following one free throw by CSU-Pueblo, the Eagles had one last chance to tie the game up with three seconds remaining. Peters ran the ball just to the outside of the arc but was blocked by Amaya Lewis as time expired. The result was a heartbreaking 77-74 loss for CSC.

Powers finished the game with 21 points and 13 rebounds, which makes for her third double-double of the season.

The Eagles will be on the road for their next two games. Friday’s game at New Mexico Highlands (5-16, 2-13 RMAC) is scheduled for a 5 p.m. tipoff and Saturday’s game at UCCS (9-12, 7-8 RMAC) will start at 5:30 p.m.

Berklee’s Signature Series celebrates Latin music in The Mambo Kings: 100 Years, with special guests including legendary percussionist John “Dandy” Rodriguez and six-time Latin Grammy Award–winning singer Gilberto Santa Rosa, on Thursday, April 6, at 8:00 p.m. at the Berklee Performance Center.

“For me it is a dream come true to bring this megastar of salsa, Gilberto Santa Rosa, to Berklee to interpret the music of Los Reyes del Mambo, Tito Puente, and Tito Rodriguez,” says Eguie Castrillo, a professor in Berklee’s Percussion Department. “Students and the general public will be traveling in a time machine back to the era of the Palladium Ballroom in New York City.”

Castrillo, the concert’s producer, will curate the music selections and lead the band, which will feature Berklee students and faculty, with Rodriguez on bongos. It will be the first time that Santa Rosa and Rodriguez have performed at Berklee.

Gilberto Santa Rosa

Nicknamed “El Caballero de la Salsa” (the Gentleman of Salsa), Santa Rosa began singing at the age of 6. Over a storied 40-plus-year career that has produced more than 25 albums, the Puerto Rican bandleader has had many firsts, including being the first tropical salsa singer to perform at Carnegie Hall. In 2021, Santa Rosa was presented with a Latin Grammy Lifetime Achievement Award, and Colegas, his most recent release, was nominated for a Grammy in the category of Best Tropical Latin Album.

John “Dandy” Rodriguez

Rodriguez is an acclaimed percussionist who has played and recorded with internationally renowned artists Tito Puente, Celia Cruz, Tito Rodriguez, La Lupe, and Johnny Pacheco, among many others. He grew up in El Barrio, known as Spanish Harlem, in New York City. At the age of 17, he earned a spot in the Tito Puente Orchestra, where he performed for over 30 years. He worked with Tito Rodriguez from 1965 to 1968, and later formed his own band, Típica 73, of which he remained a member until 1979. Rodriguez then returned to Puente’s band, playing alongside him until the time of Puente’s death in May 2000.

Admission is $15–20 in advance and $20–25 day of show. Tickets are available online and at the Berklee box office. This is a seated event.
