The Sun’s Anna Pier speaks with lifetime Sonoman Jerry Wheeler, the new manager of the Tuesday Night Market, which opens its 2022 season on Tuesday, March 3, on Sonoma Plaza.

How do things look for this year’s Tuesday market? 

I’m very excited for this year. It’s going to be a huge market. We’re back to pre-pandemic. We expect a huge turnout. It’s such a wonderful community event. People are happy when they get there. It’s an old-town gathering. Our first market is May 3.

What farmers will be there? 

We have seven large agriculture vendors, including The Patch, Mt. Moriah Farms/Smit Farms,  Neufield, Hector ‘s Honey, Triple T Ranch, Esquivel Farms, and R. Schletewitz. Seasonal vendors will be Serres Ranch, when the blueberries are ready, and Jesus’ Cut Flowers. The Patch told me they are getting ready with 5,000 tomato starts! 

Any physical changes?  

No, it will be the exact same footprint. But we have a fair amount of new vendors, and some old favorites. There were more applicants than we have space.The first priority for us is being local. This is the first year the food trucks are back since the pandemic, and we’ll have Tri-Tip Trolley and Hopmonk. Pacheco’s Roasted Mexican Street Corn is new, and we’ll have El Brinquito, E-Saan, Aunt Betty’s Gourmet Corn Dogs, Gerard’s Paella, Bruschetta Bar,  Mommy’s Yummies, Nellie’s Oysters, Kettle Pop, Java Wagon and Sweet Scoops. With packaged foods, Harvey’s Donuts, Brigachero (Brazilian truffles), Monday Bakery, the Bekjr, the Hummus Guy, Jus Verts, Sinful Sweets, Triad Beverages and Sonoma County Meats. The Chamber of Commerce will offer glasses of local wines, and Sonoma Springs will serve their beer. And there are seven artisan vendors, who have to create what they sell. All in all there will be over 50 vendors. Popo the Clown will be back with face-painting and games. 

What keeps all this in line? 

We’ve hired the Marshall Program through the Native Sons, for crowd control. And to be sure there are no dogs. We run the risk of losing our Certification if there are dogs there. Our market is certified by the California Department of Food and Agriculture. We have to be recertified annually. 

What about organic certification? 

Most of our famers have a certification of Organic Registration. That’s from the California Department of Food and Drugs. They make a site inspection. 

Music at the Market?

How did you land this spot as manager?  

I’ve always done events. I was Director of Corporate Events at the Raceway for 18 years. And I’ve coordinated the Fourth of July Parade, and I’ve been co-coordinator of the Vintners and Growers Wine Auction, and the Vintage Race Car Festival on the Plaza. 

You say this is an “old-town” market. You’re an old-town girl. 

I’ve been in Sonoma forever. My family moved up here from the Bay area in the ‘60s. We used to come up to visit my great-aunt and uncle who had a ranch on the NW corner of Grove and Carriger. Then we moved up to the top of Lovall Valley Road, and that’s where I grew up. That was way before everyone wanted to live up there.

Local schools? 

You bet. Sassarini for elementary, then 7th and 8th grades at Prestwood. That’s right, Prestwood used to be a junior high. Sonoma Valley High. I still see girlfriends that I hung out with in high school. I helped organize the Prom. 

How was it growing up in Sonoma? 

Idyllic. 

You’ve seen so many changes. 

You just have to roll with it. That’s all I can say about it. When we moved here, there wasn’t a stoplight, till they put one up at the Safeway/ Valley Mart corner. After 9PM, that one went blinking red and yellow. I remember when there really was a feed store in the Feed Store building on the corner of First St West.

Community involvement? 

I’ve worked with numerous nonprofits and other groups. I’ve served on the boards of the Chamber of Commerce, the Community Center, the Boys and Girls Clubs, Sonoma Valley Visitors Bureau, and currently Speedway Children’s Charities. 

Pastimes when you’re not working out in the community or at your other two jobs? 

I garden. My mother was a wonderful “rose tender”.  My garden – vegetables and a small rose garden – is my huge escape.

N.S. Town Hall flag raised for first time on eve of grand parade

NORTH SMITHFIELD – It’s the final touch on a building that will serve town residents for years to come, symbolically raised the evening before a massive parade set to celebrate North Smithfield’s 150th year.

Veterans with Leclair Kozlik Logan Bassett Post 6342 joined scouts from two local troops Friday, May 6 to ceremoniously place Old Glory atop a newly installed pole at 83 Greene St. A small audience watched as scouts with Troop 438 and 1139 North Smithfield hung both the American and POW flags with help from the veteran group.

The building – which once held Kendall Dean School – was fully renovated and has been used as town offices since 2020, but it was only this week that the property received the final touch.

“It’s a great occasion,” said Town Administrator Paul Zwolenski. “Tomorrow we’re going to have a parade to celebrate our 150th year.”

The town’s official anniversary of incorporation landed during March of 2021, but officials held off on planning large-scale celebrations due to pandemic concerns through most of the year.

That delay will be far from mind on Saturday as floats, marching bands, dignitaries and more line up along North Main Street. Parks & Recreation Program Coordinator Kate Pasquariello told NRI NOW last week that the event is on track to be the largest of its kind in town history.

The parade steps off from the intersection of North Main and Victory Highway at 10 a.m. and will travel along the highway, turning right on Greene Street before heading back to North Main and completing the two-mile loop. Spectator parking will be available at the former Halliwell School, Village Haven Restaurant, St. John’s Church and Pacheco Park.

While weather for Saturday’s event looked questionable for much of the week, current forecasts show the chance of rain tapering off by 10 a.m. The parade will go on rain or shine, and the flag, now flying proudly in front of Town Hall, will be there to greet it.

Protecting the human intestinal microbiome with synthetic biology

An engineered live biotherapeutic protects the intestinal microbiome against unwanted consequences of antibiotic therapies and could be developed as a simple and effective co-therapy

(BOSTON) – Many of us have experienced our gut thrown off-balance as a result of an unavoidable antibiotic therapy. Antibiotics not only kill the pathogenic bacteria causing an infection, they also indiscriminately wreak havoc on the trillions of “good” bacteria making up the human microbiota. Known as “dysbiosis,” this alteration of our gut microbial composition triggers discomforting diarrhea in up to 35% of patients in the short term, and can take months to resolve, often with the help of dietary corrections and supplements. In some patients, the microbiota can even be permanently disturbed, which becomes a serious risk factor for a host of autoimmune, metabolic, and neurological diseases.

This is because we have evolved a mutually highly beneficial relationship with the microbes in our gut microbiome and in fact formed a “superorganism” with them in which we as “hosts” shape our microbiome’s composition with our gut environment, as well as our eating and other habits. In turn, our microbial “guests” produce diverse compounds that profoundly affect the way we digest food, utilize energy, and support our immunity, among many other functions. Despite the importance of our microbiome for our health, thus far there has been no silver bullet to prevent antibiotic-induced dysbiosis.

Now, a research team at the Wyss Institute for Biologically Inspired Engineering at Harvard University and Massachusetts Institute of Technology (MIT), using a synthetic biology approach, has developed an engineered live biotherapeutic product (eLBP) that, when given together with commonly used antibiotics known as β-lactams (which includes the well-known antibiotic penicillin), protects the gut microbiome from dysbiosis. The study is published in Nature Biomedical Engineering.

“In designing the eLBP, we tapped into the synthetic biology took kit that we have advanced over the past two decades and enabled Lactococcus lactis, a safe-to-use microbe, to secrete a β-lactamase enzyme that altruistically degrades β-lactams in the bacteria’s environment,” said Wyss Core Faculty member and lead of the Institute’s Living Cellular Device Platform, James Collins, Ph.D., who led the study. “The enzyme essentially becomes a ‘common good’ that cannot confer a selective advantage to the producing bacteria or be easily transferred to other bacteria, minimizing the risk and maximizing the clinical benefits of our approach.” Collins is also the Termeer Professor of Medical Engineering & Science at MIT. In 2018, his team used L. lactis to develop an engineered probiotic intervention to detect and treat cholera infections.

Protection from antibiotics goes live

Usually, β-lactamase enzymes are encoded by a single gene that can be passed between bacteria via a process called horizontal gene transfer, and the enzymes themselves reside within the bacteria’s cell wall or membrane enclosures. This not only makes the producing bacterial strain resistant to certain antibiotics that attack these outer enclosures, but also allows their resistance to spread to other bacteria in the gut microbial population. “To safeguard against the development and spread of antibiotic resistance, we engineered various control units into the β-lactamase expression system. Essentially, we split a specific β-lactamase-encoding gene, distributed the two genetically unlinked halves to different parts of the bacterium’s DNA, and further engineered them so that they would be secreted away from the producer cell and bind to each other with high affinity to reassemble a functional enzyme in its outside environment,” said first-author Andrés Cubillos-Ruiz, Ph.D., who spearheaded the project in Collins’ group.

Cubillos-Ruiz and his co-workers on Collins’ team then showed that when they gave their eLBP intervention to mice that received the antibiotic ampicillin as an oral treatment, it indeed minimized dysbiosis in each animal’s gut. By sequencing a part of the bacterial genome known as 16S rDNA, which provides a genetic zip code for all bacterial species and families, they found that the eLBP significantly dampened the collapse of microbial populations and allowed them to fully recover their original diversity and composition three days after antibiotic treatment. Mice treated with ampicillin that were not protected by the eLPB, suffered a much greater loss of their microbial diversity which they could not recover during the entire course of the experiment.

“Importantly, during its transient stay in the digestive tract, the eLBP protected the microbiome without changing the concentration of ampicillin circulating in the blood, which is important because the antibiotic still needs to reach infections everywhere else in the body to do its job,” said Cubillos-Ruiz. “The eLBP also reduced the enrichment of various antibiotic resistance genes within the microbial community, which commonly happens under the selective pressure of antibiotics.” The gut microbiome contains a natural pool of bacteria with genes, including β-lactamase genes, that induce resistance to antibiotics through different mechanisms ­– including resistance even to unrelated antibiotics. With every antibiotic treatment their numbers shoot up and also contribute to the spread of antibiotic resistance by horizontal gene transfer.

We are now focusing on getting these living therapies to patients and are finalizing the design of an effective, short, and inexpensive clinical trial. We also believe that our general eLBP approach can be extended into a therapeutic platform that could be applied not only to other antibiotics, but also to address diseases where gut dysbiosis is at the center.

Keeping a lid on opportunistic bacteria

Finally, the team addressed a common consequence of dysbiosis: the hostile takeover of the vacated intestinal territory by problematic bacteria like Clostridioides difficile. These “opportunistic bacteria” live in many people’s intestines at lower numbers and, when given the chance to multiply uncontrollably, trigger inflammation, diarrhea, and may contribute to the development of inflammatory bowel disease. The team modeled a full-fledged human C. difficile infection by infecting ampicillin-treated mice with spores of C. difficile. The eLBP successfully prevented the intestinal colonization with C. difficile, in contrast to a normal unmodified L. lactis strain not producing the split β-lactamase enzyme.

“This is one of the strongest examples of an engineered living cellular therapy tackling a pressing clinical problem coming out of academia thus far,” said Collins.

“We are now focusing on getting these living therapies to patients and are finalizing the design of an effective, short, and inexpensive clinical trial,”said Cubillos-Ruiz, adding that, “We also believe that our general eLBP approach can be extended into a therapeutic platform that could be applied not only to other antibiotics, but also to address diseases where gut dysbiosis is at the center.”

This elegantly engineered and highly effective living cellular therapeutic device could become a true game changer in the treatment of infectious diseases both by helping to maintain a healthy microbiome in patients treated with antibiotics and, perhaps equally important in the longer run, by preventing the growing problem of antibiotic resistance which is a growing problem worldwide,” said Wyss Founding Director Donald Ingber, M.D., Ph.D., who is also the Judah Folkman Professor of Vascular Biology at HMS and Boston Children’s Hospital, and Hansjörg Professor of Bioinspired Engineering at the Harvard John A. Paulson School of Engineering and Applied Sciences.

Other authors on the study are Miguel Alcantar and Nina Donghia from Collins’ group, Pablo Cárdenas from the MIT Department of Biological Engineering, and Julian Avila-Pacheco from the Broad Institute. The study was funded by the Wyss Institute at Harvard University, a Defense Threat Reduction Agency grant (#HDTRA1-14-1-0006), and the Paul G. Allen Frontiers Group.
